After it was cooled to room temperature, Water was added dropwise to the reaction mixture under ice bath until no gas bubbles generated, and employing dilute hydrochloric acid neutralization to neutral, extracted with ethyl acetate, dried over magnesium sulfate and concentrated in vacuo to give crude product which was chromatographed on silica gel column using 1:4 to 1:2 (v/v) EtOAc-petroleum ether solution as eluent to afford isolated product beta-ketonitriles, white or light yellow solid compounds in 50% – 85% yields. Finally, stirred in concentrated sulfuric acid (3 M) at room temperature for 5 to 10 hours. The reaction mixture was neutralized to neutral by ammonia water, extracted with ethyl acetate, dried over magnesium sulfate and concentrated in vacuo to give crude product which was chromatographed on silica gel column using 1:1 to 2:1 (v/v) EtOAc-petroleum ether solution as eluent to afford isolated product beta-ketoamides 1a-p, white solid compounds in 45% – 85% yields.

The temperature is 25-30 C, the reaction is kept for 1 h, and a purple solid is precipitated in the reaction liquid. After the reaction of the raw material is completed, the reaction solution is poured into a cold aqueous solution of sodium thiosulfate to be quenched, the reaction solution is spun and filtered, and 41.7 ml is used. Recrystallization of methanol gave 20 g of 2-fluoro-5-iodobenzoic acid. The yield was 75.8% and the purity was 99.0%.

A solution of KI (2.41 g, 14.5 mmol) in water (15mL) acidified to pH 3 with a few drops of concentrated H2SO4 was added portion-wise to this boiling suspension (I2 vapors were generated with each portion). Omitting this step as in Subramanian?s reportyields the iodoso analog. The solution was filtered while still boiling, then the filtrate was stored at 4 Cfor 18 h after which time the desired compound precipitated. 2-Iodo-5-nitrobenzoic acid was thencollected by filtration while still cold to afford 1.73 g (49%) as a 13:2 mixture with the undesired2-iodo-3-nitrobenzoic acid isomer. The inseparable mixture was used as is in the next step.Characterization matches the data previously reported in the literature

Results show that the MS response of methyl iodide is a critical indicator of method robustness because of the deteriorating effects of methyl iodide on stability when sodium thiosulfate is used as an antioxidant originally described in the pharmacopoeia. With vitamin C as a newly developed antioxidant, the robustness of this method is improved significantly. A simple and sensitive gas chromatography (GC) method is described for the trace analysis of iodide anion (iodide) in processed seaweed as an organic derivative. The method is based on the derivatization of aqueous iodide extracted from seaweed with 2-(pentafluorophenoxy)ethyl 2-(piperidino)ethanesulfonate in toluene using tetra-n-hexylammonium bromide as a phase-transfer catalyst. The resulting pentafluorophenoxyethyl iodide is highly responsive to an electron-capture detector (ECD) and was analyzed by GC-ECD, giving a low detection limit of similar to2.7 nM (2.7 fmol/muL injected). Interferences of some common anions in the analysis of iodide were studied and proved to be minimal. Application of the method to the analysis of iodide in processed seaweed was performed.
