COMPARISON OF TWO IODINE QUANTIFICATION METHODS IN AN ARTIFICIAL SEAWATER SYSTEM HOUSING WHITE-SPOTTED BAMBOO SHARKS (CHILOSCYLLIUM PLAGIOSUM)

Iodine is an essential micronutrient for elasmobranchs in order to prevent goiter. Preventing goiter requires bioavailable iodide: either oral iodide or maintaining adequate aquarium water iodide concentrations. The objective of this study was to determine how oral and water supplementation affected iodine (I-2) and iodide (I) concentrations in artificial seawater aquaria housing captive white-spotted bamboo sharks (Chiloscyl hum plagiosum). Daily water samples were collected and free iodine (I-2) was determined using ultraviolet-absorbance spectrophotometry (a relatively simple in-house assay) and total iodide (I) via liquid chromatography (a more time- and expertise-intense quantification method) to learn the effects of supplementation. One water system received iodine and iodide supplementation in the form of 5% Lugol’s iodine solution added directly to the water, while a second water system received no supplementation. In addition, one tank of sharks in each water system received oral iodide supplementation. Results indicated that oral supplementation provides greater increases in water concentrations of bioavailable iodide (I) than direct water supplementation. In addition, the chromatographic results suggested that iodide is present in higher concentrations in the systems not receiving water supplementation. Increased iodide concentrations were detected in water samples after water changes and after oral iodide supplementation was administered, but total iodine (I-2) concentration changes were not detectable within the same time frame.

A novel fluorometric and colorimetric sensor for iodide determination using DNA-templated gold/silver nanoclusters

Determination of iodide ions has attracted considerable interest due to the importance and urgency. In the current work, we have developed a novel fluorescent and colorimetric platform for simple and selective detection of iodide ions based on the DNA-templated gold/silver nanoclusters (DNA-Au/Ag NCs). The detection system is based on the unique interaction between iodide ions and DNA-Au/Ag NCs, which shows an obvious fluorescence quenching with the addition of iodide ions and a good linear range from 0 to 10 mu mol/L for iodide ions. In the mean time, the detection system also exhibits an obvious color change from colorless transparent to purple red with the addition of iodide ions. Furthermore, the proposed system has been successfully applied for determination of iodide ions in real water samples.

The effects of amitrole on thyroglobulin and iodide uptake in FRTL-5 cells

Thyroid is a frequent target for endocrine effects of pesticides. Thyroglobulin (TG) and iodide uptake are crucial to thyroid hormone synthesis and may be targets of thyroid-disrupting chemicals. In our study, thyroid follicular FRTL-5 cells were treated with amitrole, an inhibitor of the thyroid peroxidase (TPO), and the effects on TG and total iodide uptake were observed. The results showed that 1 – 100 mg/L amitrole had no marked effects on FRTL-5 cell proliferation and DNA synthesis. However, it significantly increased the transcription of tg gene and inhibited the total iodide uptake. And 10 – 100 mg/L amitrole significantly decreased TG in the culture medium. The data suggests amitrole may disrupt the expression and secretion of TG and iodide uptake.
