Yang, Peng-Fei;Zhu, Lei;Liang, Jian-Xing;Zhao, Han-Tong;Zhang, Jian-Xin;Zeng, Xian-Wang;Ouyang, Qin;Shu, Wei research published 《 Regio- and Enantioselective Hydroalkylations of Unactivated Olefins Enabled by Nickel Catalysis: Reaction Development and Mechanistic Insights》, the research content is summarized as follows. Direct construction of fully alkyl-substituted tertiary chiral centers remote to activating groups is highly challenging and desirable. Herein, a Ni-catalyzed enantioselective hydroalkylation of unactivated alkenes with unactivated alkyl halides at room temperature is reported, providing a general and practical access to fully alkyl-substituted tertiary stereogenic carbon centers not adjacent to activating groups. This reaction undergoes regio- and stereoselective hydrometalation of unactivated alkenes with a nontrivial Markovnikov selectivity, followed by cross-coupling with unactivated alkyl electrophiles to access trialkyl tertiary saturated stereogenic centers not adjacent to activating groups. The mild and robust conditions enable the use of terminal and internal unactivated alkenes and unactivated primary and secondary alkyl, benzyl and propargyl halides to construct diverse trialkyl tertiary stereogenic carbon centers with broad functional group tolerance. Moreover, exptl. investigations support the reaction undergoing irreversible and stereoselective hydrometalation of alkenes. D. functional theory calculations provide further insights into the reaction mechanism, suggesting a stereoselective migration insertion of alkenes with Ni(II)-H species. Finally, the origin of the regio- and enantioselectivities was also investigated.

Chemoproteomics has enabled the rapid and proteome-wide discovery of functional, redox-sensitive, and ligandable cysteine residues. Despite widespread adoption and considerable advances in both sample-preparation workflows and MS instrumentation, chemoproteomics experiments still typically only identify a small fraction of all cysteines encoded by the human genome. Here, we develop an optimized sample-preparation workflow that combines enhanced peptide labeling with single-pot, solid-phase-enhanced sample-preparation (SP3) to improve the recovery of biotinylated peptides, even from small sample sizes. By combining this improved workflow with online high-field asym. waveform ion mobility spectrometry (FAIMS) separation of labeled peptides, we achieve unprecedented coverage of >14000 unique cysteines in a single-shot 70 min experiment Showcasing the wide utility of the SP3-FAIMS chemoproteomic method, we find that it is also compatible with competitive small-mol. screening by isotopic tandem orthogonal proteolysis-activity-based protein profiling (isoTOP-ABPP). In aggregate, our anal. of 18 samples from seven cell lines identified 34225 unique cysteines using only ∼28 h of instrument time. The comprehensive spectral library and improved coverage provided by the SP3-FAIMS chemoproteomics method will provide the tech. foundation for future studies aimed at deciphering the functions and druggability of the human cysteineome.

Xu, Xiaojia;Ji, Xiaoyu;Chen, Rui;Ye, Fangyuan;Liu, Shuaijun;Zhang, Shuo;Chen, Wei;Wu, Yongzhen;Zhu, Wei-Hong research published 《 Improving Contact and Passivation of Buried Interface for High-Efficiency and Large-Area Inverted Perovskite Solar Cells》, the research content is summarized as follows. Inverted-structured perovskite solar cells (PSCs) mostly employ poly-triarylamines (PTAAs) as hole-transporting materials (HTMs), which generally result in low-quality buried interface due to their hydrophobic nature, shallow HOMO levels, and absence of passivation groups. Herein, the authors molecularly engineer the structure of PTAA via removing alkyl groups and incorporating a multifunctional pyridine unit, which not only regulates energy levels and surface wettability, but also passivates interfacial trap-states, thus addressing above-mentioned issues simultaneously. By altering the linking-site on pyridine unit from ortho- (o-PY) to meta- (m-PY) and para-position (p-PY), they observed a gradually improved hydrophilicity and passivation efficacy, mainly owing to increased exposure of the pyridine-nitrogen as well as its lone electron pair, which enhances the contact and interactions with perovskite. The open-circuit voltage and power conversion efficiency (PCE) of inverted-structured PSCs based on these HTMs increased with the same trend. Consequently, the optimal p-PY as HTM enables facile deposition of uniform perovskite films without complicated interlayer optimizations, delivering a remarkably high PCE exceeding 22% (0.09 cm2). Moreover, when enlarging device area tenfold, a comparable PCE of over 20% (1 cm2) can be obtained. These results are among the highest efficiencies for inverted PSCs, demonstrating the high potential of p-PY for future applications.
