A series of 6-methanesulfonyl substituted benzothiazinone (BTZ) analogs I (R = N-methyl-(1-phenyl-1H-[1,2,3]triazol-4-ylmethyl)-amino, 2-phenyloxadiazol-5-yl, 4-cyclohexylmethylpiperazin-1-yl, etc.; R1 = Me, Et, i-Pr) was designed and synthesized; and further exploration introduced five-member aromatic heterocycles as linkers to attach an aryl group as the side chain. This work led to the discovery of a number of BTZ derived compounds I with potent antitubercular activity. The optimized compounds I (R = N-methyl-(1-phenyl-1H-[1,2,3]triazol-4-ylmethyl)-amino, 2-phenyloxadiazol-5-yl; R1 = Me) exhibited MIC 47 and 30 nM, resp. Compared to PBTZ169 [2-(4-cyclohexylmethyl-piperazin-1-yl)-6-methanesulfonyl-8-nitro-benzo[e][1,3]thiazin-4-one], both compounds I (R = N-methyl-(1-phenyl-1H-[1,2,3]triazol-4-ylmethyl)-amino, 2-phenyloxadiazol-5-yl; R1 = Me) displayed increased aqueous solubility and higher stability in human liver microsomes. This study suggested that an alternative side-chain modification strategy could be implemented to improve the drug like properties of the BTZ-based compds I.
