4-(4-methylpiperazin-1-yl-methyl)-3-trifluoromethyl-benzoic acid (0.755 g, 2.5 mmol, 1.0 eq), HATU (O-(7-azabenzotriazol-1-oxo)-N, N, N?, N?-tetramethyluronium hexafluorophosphate) (1.14 g, 3 mmol, 1.2 eq), DIPEA (1.29 g, 10 mmol, 4.0 eq) was dissolved in methylene chloride. The mixture was stirred at room temperature for 0.5 h and to the solution 3-iodo-4-methylaniline (582.6 mg, 2.5 mmol, 1.0 eq) was added. Then the mixture was stirred at 45 C. overnight, evaporated to dryness after the completion of the reaction, extracted with saturated aqueous sodium bicarbonate and methylene chloride 3 times. The organic layer was combined and concentrated under reduced pressure. The residue obtained was isolated by column chromatography to give the title product as a light yellow solid (892 mg, 69.0% yield). 1H NMR (400 MHz, CDCl3) 8.10 (s, 1H), 8.08 (d, J=2 Hz, 1H), 8.08-8.05 (m, 1H), 8.0 (s, 1H), 7.95 (s, 1H), 7.55 (dd, J=4, 2 Hz, 1H), 7.20 (d, J=8 Hz, 1H), 3.75 (s, 2H) 2.60-2.41 (m, 8H), 2.40 (s, 3H), 2.30 (s, 3H).
